Overview
Podoplanin, a single-transmembrane glycoprotein, has been known as a marker of lymphatic endothelium and alveolar epithelium. And also known to be overexpressed in brain tumor, malignant mesothelioma, testicular tumor, and various squamous cell carcinomas. Its expression is associated with a poor prognosis of some kinds of tumors.
Podoplanin has a highly conserved region, called PLAG domain, in its extracellular N-terminal region. In particular, podoplanin has been known to bind to CLEC-2, a receptor on the platelet, via PLAG3 domain sequence and sialylated O-glycan in PLAG3, inducing platelet aggregation. The platelet aggregation activity has been reported to be involved in metastatic and proliferation of cancer cells.
Although podoplanin shows high homology (94%) between human and monkey, this antibody does not react with monkey podoplanin, indicating a very high species specificity.
The epitope of this antibody (Gly77-Asp82) contains part of the PLAG4 domain. The PLAG4 is a newly reported domain involved in binding to CLEC-2 along with PLAG3. This antibody has a high affinity with human podoplanin.
