Anti Human Podoplanin, Monoclonal Antibody (LpMab-12)

Podoplanin, a single-transmembrane glycoprotein, has been known as a marker of lymphatic endothelium and alveolar epithelium. And also known to be overexpressed in brain tumor, malignant mesothelioma, testicular tumor, and various squamous cell carcinomas. Its expression is associated with a poor prognosis of some kinds of tumors.

Podoplanin has a highly conserved region, called PLAG domain, in its extracellular N-terminal region. In particular, podoplanin has been known to bind to CLEC-2, a receptor on the platelet, via PLAG3 domain sequence and sialylated O-glycan on PLAG3 (Thr52), inducing platelet aggregation. The platelet aggregation activity has been reported to be involved in metastatic and proliferation of cancer cells.

This antibody recognizes PLAG3 domain including its sialylated O-glycan on Thr52 of human podoplanin.

This antibody has been reported to inhibit binding between human podoplanin and CLEC-2. In particular, the sialylated O-glycan on Thr52 of PLAG3 has been known to be essential for the interaction with CLEC-2 and platelet activation.
