Anti Tau Antibodies

Tau is a microtubule-associated protein predominantly expressed in the central nervous system and is a major component of the neurofibrillary tangles found in the neurons of patients with Alzheimer's disease. Because the extent of Tau accumulation in the brain correlates with the severity of cognitive impairment, Tau is considered to play a central role in the pathogenesis of Alzheimer's disease. Diseases characterized by Tau accumulation are collectively referred to as tauopathies, and the specific isoforms that accumulate differ depending on the disease. Additionally, Tau in neurofibrillary tangles is highly phosphorylated, and this phosphorylation is implicated in the pathology of Alzheimer's disease.

What is Tau?

Tau is a microtubule-associated protein predominantly expressed in the central nervous system, where it plays a key role in regulating microtubule stability. It is a major component of the neurofibrillary tangles found in neurons of patients with Alzheimer's disease, and Tau accumulation in the brain correlates with the severity of cognitive impairment, indicating its close involvement in the development of Alzheimer’s disease1). Neurofibrillary tangles characterized by Tau accumulation are not unique to Alzheimer's disease; they are also observed in Pick's disease, progressive supranuclear palsy, corticobasal degeneration, and other conditions, which are collectively referred to as tauopathies.

In the human central nervous system, six Tau isoforms are generated through alternative splicing. These isoforms are classified into 3-repeat Tau (3R Tau) and 4-repeat Tau (4R Tau) based on the number of microtubule-binding domains (Fig. 1). In Alzheimer's disease, neurofibrillary tangles contain both 3R Tau and 4R Tau, whereas other tauopathies show disease-specific patterns of isoform accumulation. Additionally, Tau in neurofibrillary tangles is highly phosphorylated, and this phosphorylation is implicated in the pathology of Alzheimer's disease (Fig. 2).
