Product Information

Secretory IgA (s-IgA) is an IgA to which a secretory component (SC) is bound, basically occurring as a dimer but also as tetramer, etc. IgA in secretions occurs principally as s-IgA, which plays an important role in protection from infection on the mucosa of the respiratory and gastrointestinal tracts. s-IgA is also found in human saliva and produced in plasma cells in the salivary glands. The level of s-IgA in human saliva is reportedly increased by acute stress1) and decreased by chronic stress2) 3), giving it utility as a stress biomarker.

About sample

How should I store samples?
Saliva samples should be stored frozen at -30°C or below. Avoid repeated freezing and thawing of samples.
How should I prepare samples?
Dilute the saliva sample 80-fold with a buffer previously diluted with distilled water (395 μL of diluted buffer : 5 μL of saliva sample). If the saliva sample is stored frozen, thaw at room temperature and stir well. If the concentration of the diluted sample is higher than 20 μg/mL, further dilute the 80-fold diluted sample with the buffer before measurement.
How should I pretreat highly viscous saliva samples?
Highly viscous saliva may not provide a correct measurement value. Precipitate mucin, etc. by centrifugation and use the supernatant.

What reagents, instruments, and equipment are required for measurement with this kit?
The following reagents, instruments, and equipment are required to use this kit:
  • Micropipettes and disposable tips (5 to 1,000μL) (8- or 12- channel pipettes are recommended.)
  • Microplate reader which can read extinction 3.0 at 450 nm
  • Microplate shaker or conventional shaker
  • Test tubes for dilution of standard solutions and samples (glass or polypropylene)
  • Microplate washer (For manual operation, a continuous dispenser, needle dispenser, and an aspirator or vacuum pump are recommended.)
  • Graduated cylinder (1,000 mL)
  • Distilled or deionized water
