For amino acid sequencing for proteins and peptides, the Edman method developed by Edman has been widely used as a superior method. Since Edman and Begg developed the automated Edman method, the need for micro and rapid analysis has increased, so pmol-order analysis has been performed using, for example, a sequencer.Fluorescence analysis is most suitable for microanalysis by, for example, HPLC, because it is more sensitive than the ninhydrin method.It is used as a derivation agent for primary, secondary and tertiary amines, and reagents for amino acid sequence analysis by the Edman method and amino acid analysis by HPLC. It is reacted with the N-terminal amino acid in amino acid sequence analysis by the Edman degradation method. The resultant PTC derivative is treated with TFA, enriched and cut, derivatized into PTH, and amino acids identified. It reacts with an amino group containing secondary amine. It has a detection limit of 10 pmol, and the derivative is stable for 6 hours at room temperature and 3 days at 4 degrees C.
Analysis of amino acid composition is an important analysis in various fields such as protein/peptide research, food analysis and diagnosis of metabolic disorders. The PTC method, an amino acid analysis method, involves labeling with phenyl isothiocyanate (PITC), which has long been used as the Edman reagent, and is superior to other prelabeling methods in that it reacts with secondary amine and the molecular extinction coefficient for the standard compound is similar among any amino acids; however, the analysis requires complex gradients.Our company has completed a system consisting of a Wakopak WS-PTC column and a dedicated eluent allowing for analysis in short periods under simple eluting conditions. This system is applicable to any general-purpose HPLC systems supplied by other companies with few alterations in eluting conditions, and enables high-sensitivity, highly reproducible PTC amino acid analysis in short periods without troublesome consideration of conditions.Contains an ampule encapsulating nitrogen. Transfer the content to a glass-stoppered test tube after opening the ampule, and after use displace the inside of the test tube with nitrogen gas and store at -20 degrees C.
