Biochemistry
Highly Selective Fluorescent Probes
Thiol / Serenol selective fluorescent probe
Several chemiluminescent and fluorescent reagents are known as a thiol group detection reagent, and used for detection of thiol groups or measurement of cholinesterase activity.
Most of these reagents have a low hydrophilicity, so separate reaction steps, enzyme and detection reactions, are required.BES-Thio has a high hydrophilicity and can be used in aqueous solution. This feature makes it easy to measure the enzyme activity such as cholinesterase using acetylthiocholine or butyrylthiocholine as a substrate. Furthermore, by pH adjustment, BES-Thio can also detect serenol groups, which sulfur (S) in thiol group is substituted by selenium (Se), and can be used for selenoprotein detection reagent.
[Features]

  1. High hydrophilicity

  2. Respond to thiol at pH 7.4

  3. Respond to serenol at pH 5.8

Usage
  1. Prepare 5 mM ethanol solution for the Stock Solution.

  2. Probe Solution is prepared by the Stock Solution diluted with appropriate buffer be diluted by 200-fold.

  3. The probe solution can be used by mixing with Enzyme-Substrate Reaction solution, etc.

e.g.)
[Cholinesterase activity determination using 96-well microplate]
Mix 10 uL of Enzyme-Substrate Reaction solution and 170 uL of Probe Solution and incubate the solution for a certain time.
Then measure generated fluorescence.
(uL: microliters)

Detection Wavelength
Excitation wavelength: 495 nm
Emission wavelength: 535 nm
