What is laminin 511?

Laminin constitutes a part of the extracellular matrix of the basement membrane of animal cells and is involved in the processes of cell adhesion and proliferation. iMatrix-511 solution is a recombinant protein having the same sequence as laminin 511-E8 fragment. It is a cell culture substrate that promotes adhesion and extension of a variety of cells (ex. ES cells, iPS cells and mesenchymal stem cells) .

A collaborative study led by Osaka University and Kyoto University demonstrated that iMatrix-511 solution also facilitates culturing of iPS and ES cells, which was considered very difficult, in a safe and highly efficient manner.

Laminin 511 constitutes of α5-chain, β1-chain, and γ1-chain. Although laminin 511-E8 is a fragment, it has the ability to bind to α6β1 integrin as with full-length laminin.

How to use

  1. Dilute iMatrix-511 solution with PBS (-) and put onto the cell culture surface at a final concentration of 0.1-1.5 µg/cm2.
    ※The optimal concentration for coating varies with the type of cells, cell lines, and the culture medium used. For the first time usage, start with the final concentration of 0.5 µg/cm2 and determine the optimal condition for coating.
  2. Incubate in room temperature for 3 hours and discard the solution.
  3. Put cells and culture medium to start cell culture.

Feeder-free, single-cell passaging is possible when culturing ES/ iPS cells.

Example

Epidermal cells and vascular endothelial cells were cultured with iMatrix-511 solution for 0.5 and 1 hour, respectively.
