Atopic dermatitis-inducing reagent "Biostir AD"

Biostir AD is ointment including mite allergens derived from Dermatophagoides farinae. It induces atopic dermatitis in NC/Nga mice 1 to 2 weeks after application twice a week for a total of 6 times.

Induction method

After the skin barrier is destroyed with 4% SDS, Biostir AD is applied to the back and auricle of mice at a dose of 100 mg/mouse twice a week. Dermatitis is induced in approximately 3 weeks (a total of 6 doses given). A total of 600 mg of Biostir AD is applied. In this model, symptoms appear 1 to 2 weeks after the start of application, and the incidence is approximately 80% at 3 to 4 weeks after the start of application.

How to use "Biostir AD"

After barrier disruption with 4 % sodium dodecyl sulfate, apply 100 mg of Biostir AD on the shaved dorsal skin and both surfaces of each ear two times a week. Repeat this procedure for 3 weeks. The application is done 6 times in total.

1) Redness/bleeding (The back is observed for redness and bleeding symptoms.)
 0: [No symptom] Neither redness nor bleeding symptom on the back
 1: [Mild] Local redness on the back without bleeding associated with continuous scratch
 2: [Moderate] Scattered redness on the back without bleeding associated with continuous scratch
 3: [Severe] Extensive redness or bleeding associated with continuous scratch on the back

2) Crust formation/dryness (The back is observed for crust formation and dryness symptoms.)
 0: [No symptom] Neither crust formation nor dryness symptom on the back
 1: [Mild] Local crust or dryness symptoms on the back, with slightly pale skin and slight keratinous detachment
 2: [Moderate] Scattered crust or dryness symptoms, or obvious keratinous detachment on the back
 3: [Severe] Extensive crust or obvious keratinous detachment on the back

3) Edema (The auricle is qualitatively observed for edema.)
 0: [No symptom] No thickening of either auricle
 1: [Mild] Slight thickening of either auricle
 2: [Moderate] Obvious thickening and tightness of both auricles
 3: [Severe] Obvious thickening, tightness, and curvature of both auricles with hardness when touched with a finger

4) Scratch/tissue loss (The auricle is qualitatively observed for scratch and tissue loss symptoms.)
 0: [No symptom] Neither scratch nor tissue loss symptom in the auricle
 1: [Mild] Non-continuous scratch in the auricle without tissue loss
 2: [Moderate] Small continuous scratch in the auricle without tissue loss
 3: [Severe] Continuous scratch and tissue loss in the auricle
