Jackson社 金コロイド標識体
Jackson社の金コロイド標識体は、３つのグレードを用途別にラインアップしています。
EM(Electron Microscopy)グレード：透過電子顕微鏡(TEM)、走査電子顕微鏡(SEM)用
EMグレードの金ナノ粒子は、密度勾配遠心分離により小凝集塊から丹念に分離した単量体粒子です。この単量体金コロイド-タンパク質複合体は、金コロイド粒子の大きさにより異なる抗原部位を識別することができることから、多重染色に適しています。金コロイドの大きさによって、6nm、12nm、18nmのラインアップがあり、全ての製品は、安定剤と保存剤入りの滅菌濾過済みバッファー中に懸濁しています。 粒子径が4nm以上の金コロイドを光学顕微鏡やイムノブロッティングに使用したい場合も、本品をご使用ください。
LM(Light Microscopy)グレード： 光学顕微鏡、イムノブロッティング用
小凝集塊を除去していない金コロイド-タンパク質複合体で、粒子径の変動係数は15%以下です。LMグレードの金コロイドの大きさは4nmで、粒子が小さく組織へ浸透しやすいことから、光学顕微鏡を用いた銀増感法に適しています。安定剤と保存剤入りのバッファー懸濁液を凍結乾燥しています。
40 nm ImmunoGold： Lateral flow Immunoassays (LFIAs)用
40nmの金コロイド複合体です。目視可能な強い呈色を示す上、その結合が単純であることから、ラテラルフローイムノアッセイ(LFIAs)に適しています。二次抗体以外にも、コントロールとして Chrompure™ purified proteins に40nm金コロイドを標識した製品のラインアップがあります。
EM(Electron Microscopy)グレード［透過電子顕微鏡(TEM)、走査電子顕微鏡(SEM)用］
二次抗体に単量体金コロイドを標識したものです。電子密度の高い金は、透過電子顕微鏡（TEM）や走査電子顕微鏡（SEM）において電子散乱を高めるため、強いコントラストを持つ「点」として観察されます。異なる大きさの金コロイドを標識した二次抗体を使用することで、多重染色に使用できます（図1）。透過電子顕微鏡（TEM）や走査電子顕微鏡（SEM）に加え、光学顕微鏡にも使用可能です。

[固定したヒト角膜切片上のMUC16とezrinの共局在]
固定したヒト角膜切片の微絨毛上にあるMUC16（12 nm Colloidal Gold AffiniPure Donkey Anti-Mouse IgG (H+L))[メーカーコード：715-205-150]とezrin（6 nm Colloidal Gold AffiniPure Donkey Anti-Goat IgG (H+L)）[メーカーコード：705-195-147]の共局在を透過電子顕微鏡（TEM）で示した。

ヒト角膜上皮培養細胞上のMUC16 タンパク質の局在化
重層化したHCLE培養細胞中のMUC16 タンパク質の位置をMonoclonal Mouse Anti-Human MUC16 antibody(OC125; Dako)と12 nm Colloidal Gold AffiniPure Donkey Anti-Mouse IgG (H+L)[メーカーコード：715-205-150]を用いて特定した。

金コロイドの大きさが4nmのLMグレードは、粒子が小さいため、優れた組織浸透性を示します。光学顕微鏡を用いた銀増感法に適しています。
HRP標識二次抗体と金コロイド標識 抗-HRP抗体を使用した拡散光の少ない組織画像の取得
HRP標識二次抗体に金コロイド標識 抗-HRP抗体を結合させ、銀増感試薬を用いると、HRP標識二次抗体とHRP基質(diaminobenzidine:DAB)の検出系より、拡散光の少ない組織画像を得ることができます。

40 nm ImmunoGold［Lateral flow Immunoassays (LFIAs)用］
ラテラルフローイムノアッセイ(LFIAs)法を利用した迅速検査手法は、幅広い分析対象物質の検出や測定を素早く行う手段として、昨今非常に注目を集めています。40nm金コロイド複合体である本製品群は、強い呈色を示す上、その結合が単純であることから、ラテラルフローイムノアッセイ(LFIAs)法に適しています。
