Outline Resveratrol is a selective inhibitor of cyclooxygenase-1 (COX-1) (ED50=15 μM). It inhibits hydroperoxygenase activities of COX-1 (ED50=3.7 μM).

Histone deacetylase (HDAC) plays a central role in chromatin structure formation associated with the nuclear distribution of DNA. There are presently 17 known types of this enzyme in mammals, which are classified into 3 classes (see below). Also, HDAC Class III has been reported to be associated with regulation of aging and life span.


  • Mammalian HDAC Class I: Localized in the nucleus. Similar to RPD3 enzyme. [Types: HDAC1-3, 8]

  • Mammalian HDAC Class II: Localized in the nucleus and cytoplasm and shuttles between the two. Similar to HDAC1 enzyme. [Types: HDAC4-7, 9, 10]

  • Mammalian HDAC Class III: NAD-dependent enzyme. Localized in the nucleus and cytoplasm. Also called Sirtuin and is similar to SIR2 enzyme. [Types: Sirt 1-7]


Resveratrol, which is a polyphenol found in red wine, lowers the Michaelis constant of SIRT1 for both the acetylated substrate and NAD(+), and increasescell survival by stimulating SIRT1-dependent deacetylation of p53. In yeast, resveratrol mimics calorie restriction by stimulating Sir2, increasing DNA stability and extending lifespan by 70%1).
