Kv2.1/Kv2.2 Channel Blocker
Enhancer of Glucose-Dependent Insulin Secretion


Pancreatic β cells respond to glucose, close the ATP-sensitive K+ channel, and increase intracellular Ca2+ to cause insulin secretion. β cells have delayed rectifier K+ channels that play a major role in repolarizing the action potential. Inhibition of these channels increases intracellular Ca2+, causing enhanced insulin secretion. Therefore, delayed rectifier K+ channel blocker has a definite advantage over ATP-sensitive K+ channel blocker in the treatment of type 2 diabetes. Kv2.1 channel, which is a delayed rectifier K+ channel, expresses markedly in β cells. As a result of screening 85 toxicants by an assay system with 86 Rb+ using CHO cells expressed Kv2.1 channels, Guangxitoxin-1E (GxTX-1E, not more than 1 nM of IC50) was found in the Chinese venomous spider, Plesiophrictus guangxiensis sp. nov.
