Title: In-depth proteome profiling of EVs for quality assessment and liquid biopsy development

Numerous studies on EVs in recent years have led to great interest not only in elucidating their basic biological functions, such as biosynthetic pathways and functions, but also as diagnostic and therapeutic tools. In fact, liquid biopsy targeting EV-encapsulated molecules and therapies using MSC-derived EVs are approaching practical application. On the other hand, the key to any EV research is the purity of the EVs to be handled. For example, it is not difficult to imagine that the purity of the EVs will greatly affect the results of biomarker discovery based on omics analyses of plasma-derived EVs or those of MSC-derived EV therapies. In this presentation, we will show the results of our EV enrichment score (EES) evaluation method based on in-depth proteome analysis and the evaluation results of various EV purification techniques. Based on these assessments, we will also show the results of our research for the development of biomarkers for cancer or dementia using EVs purified by the MagCapture Exosome Isolation Kit PS Ver. 2 (FUJIFILM Wako Pure Chemical Corporation) on the 96-well processing robot. We will also introduce the MassivEV EV Purification Column PS for mass production of therapeutic EVs and EV-Save reagent that minimizes adsorption loss during storage. These tools are expected to further accelerate basic research and clinical applications of EVs.
