Leaf discs shredded and enzyme-treated still contain a lot of tissue debris, and thus centrifugal settling is not sufficient for protoplast purification. Utilizing the nature of protoplasts that can be suspended in a sucrose solution, broken protoplasts are separated and purification is accomplished. By adding Ficoll or Percoll into the sucrose solution, the suspension of protoplasts occurs more easily.
