Endotoxin is a component of the outer membrane of Gram-negative bacteria and is a lipopolysaccharide. Since endotoxin causes serious symptoms such as fever and shock when it comes into a human body, the endotoxin test for pharmaceuticals, especially parenteral drugs, is important. Since Levin and Bang discovered that endotoxin gels with an extract from horseshoe crab amebocytes (Limulus amebocyte lysate, or LAL), endotoxin detection by the Limulus test has been widely used. However, the specificity of LAL has been an issue recently. It was reported that LAL reacted with not only endotoxin but also beta-1,3-glucan, and that there was another cascade system in LAL activated by beta-1,3-glucan other than that activated by endotoxin. It was also reported that an LAL-reactive material was eluted from cellulose membranes, which interfered with endotoxin assays. Research on endotoxin-specific LAL revealed that LAL activation by a glucan in a sample was inhibited by a sufficient amount of CM-curdlan, added to the LAL. It was found that endotoxin assays were not interfered with by beta-1,3-glucan if a large amount of CM-curdlan. An endotoxin-specific LAL will be prepared when LAL ES-2 is reconstituted with endotoxin-free water.
