Outline

Embedding involves allowing the tissue specimen (mass) to have given uniform hardness and filling the hollows in the tissue with an embedding agent such that the tissue is hard enough to resist deformation during sectioning. It also increases preservability such as ease of handling.
Generally, embedding with paraffin is used for light microscopy and embedding with various resins for electron microscopy; the freezing method has been increasingly used in place of embedding.

Various resins including epoxy resins are used as embedding agents for electron microscopy to meet the following requirements.
- Not contracting upon polymerization.- Monomers are easy to handle and penetrate into tissues.- Clear and preferably colorless.- Polymerized resins are resistant to electron beams.- Polymers of appropriate hardness can be prepared.- Easy to section.Epoxy resin embedding materials are adjustable for hardness by altering the type and amount of addition of a hardening agent, polymerization catalyst, accelerator, etc.
Main ingredient: Ethylene glycol diglycidyl ether.
