Amino acid analysis by the OPA (o-phthalaldehyde) method is higher in detection sensitivity than the ninhydrin method. The method can analyze an amino acid of as low as 10 pmol, and a dedicated automatic analyzer for OPA has been widely used. The product is a set of OPA reagents suitable for use with the automatic amino acid analyzer, and can be prepared readily by mixing 2 solutions in equal quantities and used simply by installing into the equipment.

Metabolism of sympathetic catecholamine plays a key role in circulatory diseases (ischemic diseases, cardiac failure, hypertension, etc.). The high-performance liquid chromatography is used for analysis of catecholamine with detection by the ninhydrin coloring, OPA-fluorescence method and electrochemical determination (ECD).
Measurement principle
o-phthalaldehyde method - principle and structure
Under alkaline conditions, reaction in the presence of 2-mercaptoethanol at 50 °C for no more than 1 minute will result in generation of fluorescent material several-fold more intense than fluorescamine. The reagent is relatively inexpensive, stable in water and may be stored in solution.

OPA Reagent Set

Kit Contents:

  1. OPA Solution, containing 0.14 w/v% o-Phthalaldehyde ......
    500 mL x 2 bottles

  2. 2-ME Solution, containing 2-Mercaptoethanol ......... 500 mL x 2 bottles

Usage:
Equal amount of OPA Solution and 2-ME Solution is mixed together to make the working solution just before use.

For proline and hydroxyproline analysis, add 10% sodium hypochlorite solution at the rate of 1 ~ 10 mL per 1 L of OPA Solution. Even in this case, it in unnecessary to add extra 2-ME Solution.
The working solution should be used on the same day.

Note:

  1. Prepared working solution can be kept for one week in a refrigerator.

  2. The working solution containing sodium hypochlorite for proline and hydroxyproline analysis should be used up on the same day.

  3. Ammonia in an eluent mayl elute along with lysine and ammonia in your sample
    trapezoidally. So, the baseline of this area will rise.

  4. There is no need to intriduce nitrogen gas.
