Overview / Applications

Outline This is a reagent that introduces an electron-attracting substituent to the carboxyl group of an amino acid or peptide for activation in order to bind the carboxyl group to the amino group of amine components. Those with an activation substituent introduced to the carboxyl group are called active esters and are generally susceptible to moisture absorption in many cases, but can be isolated and purified and withstand long-term storage in some cases. They are used for increasing the yield of reactants as well as for isolation.The C-teminal of peptide and amino acid can be activated by introducing an electron-attracting substituent to the C-terminal. N-hydroxysuccinimide (HOSu) ester, like pentafluorophenyl (OPfp) ester, has this C-terminal activation action and is called an active ester. This active ester has advantages of mild reaction, generally reacting only with the amino group, not causing transesterification side reaction, and capablity to be isolated.HOSu active ester may be used being isolated but like HOBt, may be often used as an additive for reaction with DCC.
