| Outline | <Enzyme Substrate> It is a substrate of 5-aminolevulinate dehydratase and is widely distributed in various living organisms. Usually, it is synthesized by 5-aminolevulinate synthase from succinyl CoA and glycine via 2-amino-3-oxoadipinate. Some patients with porphyria show synthesis of a large amount of aminolevulinic acid in the liver. For treatment of this disorder, administration of aminobenzoate to divert glycine to the synthesis of hippuric acid and its amino derivaties is said to be effective to reduce the amount of aminolevulinic acid synthesized. 5-aminolevulic acid is an unstable compound of which two molecules form porphobillinogen through condensation. |
