Dialysis is a technique that uses a semipermeable membrane to allow low-molecular-weight solutes to diffuse naturally from a high-concentration solution to a low-concentration solution until equilibrium is reached (Figure). This process reduces the concentration of low-molecular-weight substances present in the sample. Dialysis is a simple and gentle method for removing low-molecular-weight components and is used in a variety of experimental applications, as described below.

  • Protein concentration

  • Buffer exchange

In dialysis procedures, regenerated cellulose membranes are commonly used because they exhibit low protein adsorption and high chemical and thermal stability. Molecules smaller than the membrane pore size can pass through the membrane, whereas larger molecules are retained; therefore, selecting an appropriate pore size allows selection of substances to be retained based on molecular weight. The molecular weight cut-off (MWCO), used as a guideline, represents the molecular weight at which 90% or more of the substance is retained. However, because permeability is also influenced by factors such as solute shape, degree of hydration, ionic charge, and polarity, it is recommended to select an MWCO that is less than half the molecular weight of the substance to be retained.
