CHIR99021

CHIR99021 is a highly selective GSK-3 (glycogen synthase kinase 3) inhibitor. It shows no cross-reactivity with CDKs (cyclin-dependent kinases).It is used in differentiation induction processes to generate neural cells, cardiomyocytes, hepatocytes, retinal cells, pancreatic islet β-cells, renal tubular cells, and nephron progenitor cells from human ES/iPS cells.

Overview

CHIR99021 is a potent glycogen synthase kinase 3 (GSK3) inhibitor and a compound classified as an aminopyrimidine derivative. It inhibits both GSK3β (IC₅₀ = 6.7 nM) and GSK3α (IC₅₀ = 10 nM) (Ring D, et al., 2003).

GSK3 inhibitors suppress the activity of GSK3 within the Wnt/β-catenin pathway and promote the nuclear translocation of β-catenin by inhibiting its degradation. Since Sato et al. demonstrated in 2004 that activation of the Wnt pathway via GSK-3 inhibition plays a crucial role in maintaining the pluripotency of mouse and human embryonic stem cells (Sato N, et al., 2004), there has been active development of inhibitors and activators related to this signaling pathway. Among these, BIO, SB-216763, and CHIR98014 are known as representative GSK-3 inhibitors; in particular, CHIR99021 is characterized by low cytotoxicity and high ability to activate the Wnt/β-catenin pathway.

CHIR99021 is used for a wide range of applications, including somatic cell reprogramming, maintenance of pluripotent stem cells, promotion of cell proliferation, and even the induction of differentiation into specific tissue cells.

Key Applications of CHIR99021

Maintenance and Self-Renewal of Stem Cells

  • When used in combination with PD0325901 in the presence of LIF, CHIR99021 promotes the maintenance of undifferentiated mouse ES cells (Ying QL, et al., 2008)。
  • CHIR99021 promotes the proliferation of mouse and human intestinal stem cells(Wang F, et al.,2015
  • CHIR99021 promotes the proliferation of NOD mouse-derived ES cells when used in combination with Wnt3a and Kenpaullone(Hanna J, et al, 2009)。

Reprogramming

  • CHIR99021 enables the chemical reprogramming of mouse embryonic fibroblasts when used in combination with Forskolin, Tranylcypromine, Valproic Acid, 3-Deazaneplanocin A, and E-616452(Hou P, et al.,2013)。
  • Induction of pluripotent stem cells is possible from non-responsive mouse strains(Garbutt TA et al., 2020
  • When combined with OCT4 and other small molecules, CHIR99021 promotes the reprogramming of human somatic cells into iPS cells(Zhu S, et al., 2010)。
  • In combination with Forskolin, ISX-9, SB431542, and I-BET151, CHIR99021 enables direct reprogramming of mouse fibroblasts into mature neurons(Li X, et al., 2015)。

Differentiation Induction

  • CHIR99021 promotes the differentiation of human iPS cells into insulin-producing cells.(Kunisada Y, et al., 2012
  • CHIR99021 promotes the differentiation of human ES cells and iPS cells into cardiomyocytes.(Lian X, et al., 2012
  • CHIR99021, when used in combination with SB431542 and human recombinant LIF, induces and maintains neural stem cells derived from human embryonic stem cells.(Li W, et al., 2011
