Creatinine Assay Kits

Creatinine is a metabolic product formed by the dehydration and cyclization of creatine and is a type of non-protein nitrogen. The produced creatinine is excreted in the urine via the kidneys. The serum creatinine level rises when kidney function is compromised.

LabAssay™ Creatinine is a kit used for the determination of creatinine in blood (serum, plasma) and urine using the Jaffé reaction. With the use of a microplate, this kit provides a convenient method for measuring creatinine in samples.

What is Creatinine?

Creatinine is a metabolic product formed by the dehydration and cyclization of creatine and is a type of non-protein nitrogen. Phosphorylated creatine (creatine phosphate) is an important energy source for muscle contraction, and creatinine, a metabolic product of creatine, is produced mainly in the muscles. In humans, the daily conversion rate from creatine to creatinine is almost constant. Unlike the biochemically similar test item, blood urea nitrogen (BUN), the amount of creatinine in the serum is not affected by protein intake. On the other hand, muscle mass and the level of exercise influence creatinine production1).

The produced creatinine is excreted in the urine via the kidneys. The serum creatinine level rises when kidney function is compromised. The creatinine level increases significantly when about 70% of the nephrons in both kidneys become dysfunctional. Conversely, the serum creatinine level often remains within the normal range in cases of mild to moderate kidney dysfunction, making it an inappropriate indicator for early kidney dysfunction.

Methods of Creatinine Measurement

LabAssay™ Creatinine is a creatinine measurement kit that utilizes the Jaffé reaction. The Jaffé reaction is widely used for creatinine measurement because of its simple procedures and a high degree of coloration2). On the other hand, substances with active methylene groups require caution, because they are Jaffé reaction-positive substances (non-creatinine chromogens) and affect the measurement values.

Principle of the Creatinine assay using Jaffé reaction

A protein remover solution is added to the sample and centrifuged, and the supernatant is obtained. When picric acid is added to the supernatant, the creatinine in the sample reacts with the picric acid in an alkaline solution to produce an orange-red condensation product (Jaffé reaction). By measuring the absorbance of this orange-red color, the creatinine concentration in the sample is determined.
