Protocol
Preparation of 1% Agarose S gel
- Add one tablet to 50 mL of TAE or TBE buffer, leave for 5 minutes at room temperature.
- Leave for 5 minutes at room temperature to allow the tablet to fall apart. *1
- Shake gently and heat in a microwave oven or the like after confirming there is no solid material.
Usage Note
Do not heat the solution before the tablets have fallen apart.
When heated in the presence of solid materials with a microwave oven, a gel film is formed on the surface of the solid materials preventing further melting of Agarose, as shown in the bad example below.

Effect of Agarose concentration on the migration distance of bands
Molecular weight markers were electrophoresed using Agarose S 0.5 % gel, 1 % gel and 2 % gel, and the effect of the Agarose gel concentration on the migration distance of the band was investigated. (Use as a reference for determining the concentration when a gel is prepared.)
