Histochemical staining generally requires a separate tissue section for each staining method. If stained tissues can be decolorized and re-stained, there is no need for multiple sections, and highly accurate clinical examination and research can be performed. Fujifilm Wako offers the novel tissue decolorizing reagents "deColorizing Solution." This reagent allows decolorization of Hematoxylin and eosin (H&E) or Masson trichrome (MT)-stained tissue sections for re-staining with other staining methods without damaging the tissue.

Challenges in Histochemical Staining and Novel Reagents for Decolorizing Tissue Specimens

Histochemical staining generally requires a separate tissue section for each staining method. However, depending on the size and condition of the specimen, it may be difficult to prepare multiple tissue sections. Sometimes, the structure to be observed may not span multiple (adjacent) tissue sections.

In such cases, if stained tissues can be decolorized and re-stained, highly accurate clinical examination and research can be conducted. In particular, hematoxylin and eosin (H&E) staining is frequently used as the first staining method, and there has been a need for a technique that can decolorize and re-stain H&E-stained tissues.

In the past, hydrochloric acid in ethanol or potassium permanganate-oxalic acid solutions have been used to decolorize stained tissues. However, the hydrochloric acid in ethanol was insufficient to decolorize hematoxylin, while potassium permanganate-oxalic acid solutions caused significant damage to the tissue sections.

Fujifilm Wako offers a novel tissue decolorizing reagents, deColorizing Solution, developed by Drs. Aisa Ozawa and Motoharu Sakaue of Azabu University. This reagent allows decolorization of H&E- or Masson’s trichrome (MT)-stained tissue sections for re-staining with other staining methods without damaging the tissue (Figure 1).
