Introduction

Creatinine is a metabolite which is produced by creatine phosphate directly or dehydration of creatine in muscle and nerve. It is excreted from the body through kidney glomerular filtration.

Assay principle

LabAssay™ Creatinine can be used to measure the creatinin levels in samples by Jaffe's reaction where creatinine produces quantitatively an orange color with picric acid in alkaline condition.

Procedure

Perform the assay in the wells according to the following table scheme.

First operations should be performed in the sample tubes.

Mix well, and stand at room temperature for 10 min.
Centrifuge over 2500 rpm for 10 min.
Subsequent operations should be performed in the wells.

  Stand at 25 ℃ - 30 ℃
Picric Acid Reagent 50 μL 50 μL 50 μL
0.75 mol/L Sodium Hydroxide Solution 50 μL 50 μL 50 μL
  Stand at 25 ℃ - 30 ℃ for 20 min.
Measure the absorbance of the test sample and standard solution
at 520nm with the blank solution as the control within the next 30 min.
