Product Information
Chromogranin A (CgA) is an acidic glycoprotein that coexists and is co-released with catecholamine in the adrenal medulla and sympathetic nerve endings. CgA is thought to reflect secretion of catecholamine and is used as an activity index for the sympathetic-adrenal-medullary (SAM) axis, a stress-response pathway. It is also used as a tumor marker for patients with pheochromocytomas or neuroendocrine tumors, who have high levels of CgA in the blood.
CgA is also present in saliva, is synthesized in the submandibular gland, and is released into saliva in response to autonomic stimulation. Previous studies have reported that saliva CgA levels increase in response to stress1) 2), and thus, it is particularly useful as an excellent noninvasive biomarker for mental stress.
Human Chromogranin A ELISA Kit Wako is a competitive ELISA kit for assaying CgA in human saliva and plasma.

FAQ
About sample
- How should I store samples?
- Plasma and saliva should be measured immediately after collection. If measurement cannot be performed immediately, divide the sample into small aliquots and store frozen at -30°C or below. Avoid repeated freezing and thawing of samples.
- Which anticoagulant should I use for blood samples?
- EDTA plasma, heparin plasma, or citrate plasma can be used. The same anticoagulant should be used throughout the measurement.
- How should I pretreat highly viscous saliva samples?
- Highly viscous saliva may not provide a correct measurement value. Precipitate mucin, etc. by centrifugation and use the supernatant.
About analyte
- What is the degree of cross-reactivity with chromogranin A processing products?
- It shows 100% cross-reactivity with the synthetic N-terminal fragment of parastatin [human CgA (356-374)]. It does not cross-react with pancreastatin or β-granin at all.
Fujifilm Wako have not checked cross-reactivity with vasostatin or catestatin, but based on the antibody recognition site (human CgA344-374) of the product, its cross-reactivity with vasostatin is expected to be very low, while cross-reactivity with catestatin is possible.
- What is the degree of cross-reactivity with non-human species?
- It shows 70% cross-reactivity with rat chromogranin A.
Fujifilm Wako have not checked cross-reactivity with other species.
About the kit
- What is the origin of the standard?
- The origin is synthetic human chromogranin A (344-374).
- Can I divide plates?
- Since the plate in this kit are separable and each row (8 wells) is detachable, divided use of the kit is possible.
About kit usage
- What reagents, instruments, and equipment are required for measurement with this kit?
- The following reagents, instruments, and equipment are required to use this kit:
- Micropipettes and disposable tips (25 to 1,000μL) (8- or 12- channel pipettes are recommended.)
- Microplate reader which can read extinction 2.5 at 490 nm
- Microplate shaker or conventional shaker
- Test tubes for dilution of standard solutions and samples (glass or polypropylene)
- Microplate washer (For manual operation, a continuous dispenser, needle dispenser, and an aspirator or vacuum pump are recommended.)
- Graduated cylinder (1,000 mL)
- Distilled or deionized water
