Outline Amino Acid Analysis Reagents are high-purity reagents from which ninhydrin positive components that would cause interference in the analysis have been removed.Ninhydrin reacts with amino acids and yields the bluish purple Ruhemann's purple at around pH 5. For this reaction, reduced ninhydrin needs to be formed first. Therefore, reducing agents such as tin chloride or titanium chloride are added to the ninhydrin reagent for quantitative analysis of amino acids, or reduced ninhydrin (hydrintantin) is employed with the reagent. Methyl cellosolve is usually used as the solvent for ninhydrin reagent, however, as methyl cellosolve is toxic, it is sometimes replaced by dimethyl sulfoxide. As ninhydrin solution is easily oxidated in the air, it is prepared under nitrogen.Ninhydrin reagent is said to be stable for about 2 weeks. However, since its color development decreases gradually with time, a certain amount of an amino acid other than the constituent amino acids in the analyte protein should be added as an internal standard, and the analytical value should be corrected using the recovery rate of the internal standard. The detection limit is 100 pmol maximum, and 250 pmol for proline.
