Overview / Applications

Outline Nitric acid for fine chemistry has a grade equal to or higher than special grade, and is used for sample pretreatment using plenty of acids, low background in colorimetric analysis, and high sensitivity instrumental analysis.
In preparing thin sections, the presence of bone tissue, calcification and calcinosis will hinder the preparation of desirable sections. The decalcification process is therefore required. The process involves decomposing calcium carbonate in tissues with acids into soft calcium salts to facilitate the preparation of thin sections. Decalcification agents must meet the following requirements.- Decalcification is completed readily- Decalcification does not cause tissue deformations such as swelling, contraction and lysis.- Tissue stainability is not affected- The process requires operations that are as simple as possible and inexpensiveNitric acid solution in the concentration range 5 to 10 % is most convenient. Decalcification is completed in about 10 to 20 minutes in bone marrows including trabeculae of bone and kidneys containing sand-like lime contents, and mostly in about 1 day. Relatively good stainability is preserved. However, a too long time of decalcification will inevitably result in some adverse effects on stainability.
