Outline In the peptide bond formation reaction, amine component and acid component are protected by a temporary protective group and a semi-permanent protective group. For extension of peptide bonds, temporary protective groups such as Boc group are eliminated selectively, and the next peptide chain is extended. It is important to select the correct combination of the protecting group and the deprotecting group because the protecting group to be eliminated differs depending on the type of deprotecting reagent.It is used in the preparation of Fmoc- amino acids by introducing an Fmoc group to the N terminal of amino acids.
Fmoc- has become utilized as an amino group protector instead of Boc- and Z- (especially in the solid-phase synthesis) in the peptide synthesis .
