Outline	Fluorescence analysis is most suitable for microanalysis by, for example, HPLC, because it is more sensitive than the ninhydrin method.It reacts with amino groups including secondary amines, thiol group, phenol group and imidazole group. The detection limit is less than 1 pmol . The derivatives are stable at room temperature for 30 hours (excluding Fmoc amino acid).In the peptide bond formation reaction, amine component and acid component are protected by a temporary protective group and a semi-permanent protective group. For extension of peptide bonds, temporary protective groups such as Boc group are eliminated selectively, and the next peptide chain is extended. It is important to select the correct combination of the protecting group and the deprotecting group because the protecting group to be eliminated differs depending on the type of deprotecting reagent.It is used in the preparation of Fmoc- amino acids by introducing an Fmoc group to the N terminal of amino acids.Wavelength for measurement: [Lambda]ex = 264 nm, [lambda]em = 340 nm
