Data
Data1. Approximate RNA yield
| Plant | RNA yield |
|---|---|
| Spinacia oleracea (leaves) | 0.5 μg RNA/mg tissue |
| Brassica oleracea var. italica (seedlings) | 1.0 μg RNA/mg tissue |
| Brassica oleracea var. italica (seeds) | 1.3 μg RNA/mg tissue |
| Brassica oleracea var. capitata (seedlings) | 1.4 μg RNA/mg tissue |
| Brassica oleracea var. capitata (leaves) | 0.1 μg RNA/mg tissue |
| Bambusoideae (leaves) | 0.3 μg RNA/mg tissue |
| Tulipa gesneriana (bulbs) | 0.2 μg RNA/mg tissue |
Data2. RNA purification from Arabidopsis thaliana leaves
RNA is purified using “ISOSPIN Plant RNA” and “Competitor A”. The quality of the extracted RNA was evaluated by measuring the RNA Integrity Number (RIN) using Bioanalyzer (Agilent), performing electrophoresis, and measuring absorbance.
| Kit | A260/A280 | A260/A230 | RNA yield (ng/mg tissue) |
RIN* |
|---|---|---|---|---|
| Competitor A | 1.95 | 1.13 | 72 | 7.8 |
| ISOSPIN Plant RNA | 2.35 | 2.69 | 79 | 7.5 |
*N=2

Lane① : RNA extracted using Competitor A
Lane② : RNA extracted using ISOSPIN Plant RNA
RNA (100 ng) are loaded in each lanes.
Result
ISOSPIN Plant RNA extracted higher-purity RNA compared to Competitor A. In electrophoresis, the RNA extracted using ISOSPIN Plant RNA showed a stronger signal than that from Competitor A.
Data3. RNA purification from leaves and seeds of Brassica oleracea var. capitata
RNA was extracted from the leaves and seeds of Brassica oleracea var. capitata using ISOSPIN Plant RNA and Competitor A."
1. Absorption Spectrum Measurement

2. Electrophoresis

[RNA Volumes]
Leaves 1 μg
Seeds 0.5 μg
[Marker]
RNA ladder
Denatured by formaldehyde 1% Agarose S
3. Real-Time qPCR

Result
The absorption spectrum results indicate that ISOSPIN Plant RNA more effectively removes polysaccharides and other contaminants compared to Competitor A, as the absorbance of RNA extracted using ISOSPIN Plant RNA was lower than that of RNA from Competitor A."
Data4. Improved RNA purity by using “Assist Buffer for ISOSPIN Plant RNA”
RNA was extracted from three plant species that are difficult for RNA extraction, using “ISOSPIN Plant RNA” alone or “ISOSPIN Plant RNA” with “Assist Buffer for ISOSPIN Plant RNA”.

Result
Using “ISOSPIN Plant RNA” with “Assist Buffer for ISOSPIN Plant RNA” resulted in higher RNA yield and purity compared to using ISOSPIN Plant RNA alone.
